solution, which prompted an immediate discharge from the cutaneous granular glands. The skin was then cut into small pieces and cultured in a nutrient/salts medium in a manner similar to that described by Baily (1971). One of the amino acids in the medium was replaced by the 3H-labelled amino acid. The skin cultures were incubated for up to 7 days at 28°C. After 3 and 7 days, samples of skin were first treated with an adrenaline solution, then dried in vacuo and extracted with methanol. The adrenaline solution, the methanol extract and the remaining medium were examined for the presence of jH-labelled caerulein. Thesampleswere mixed with an authentic sample of caerulein and examined by paper electrophoresis or paper chromatography. Samples from the adrenaline solution and the methanol extracts after 3 or 7 days incubation gave no radioactive peaks corresponding to the caerulein marker. However, samples of the medium in which the skins had incubated for 3 days gave a small peak of radioactivity which co-migrated with caerulein. When this solution was kept at 37°C over a period of several hours it was found that the amount of radioactivity which co-migrated with caerulein increased considerably. This suggests that caerulein matures during this period from a precursor molecule.

Membrane lipid physical state and modulation of the Na+,Mg2+-ATPase activity in Acholeplasma laidlawii B.
Careful analysis of the Arrhenius plot of the Na+,Mg2+-ATPase (ATP pyrophosphohydrolase, EC 3.6.1.8) activity in Acholeplasma laidlawii B membranes of varying fatty acid composition has been combined with differential thermal analysis of the membrane lipid phase transitions to evaluate the effects of membrane lipid properties on the enzyme activity. Our results indicate that the enzyme is activ...
متن کاملFusion of phospholipid vesicles with viable Acholeplasma laidlawii.
When vesicles of dipalmitoyl phosphatidylcholine produced by sonication are mixed with Acholeplasma laidlawii in neutral buffer, a phenomenon occurs that is most readily explained in terms of fusion of lipid vesicle membrane with the mycoplasma cytoplasmic membrane. The mycoplasma can readily accumulate large quantities of the foreign lipid without loss of viability. The added lipids do not rem...
متن کاملEvidence for two pools of cholesterol in the Acholeplasma laidlawii strain B membrane: a deuterium NMR and DSC study.
Recent investigations have indicated that there exists a well-defined range of membrane hydrocarbon order compatible with good growth of the microorganism Acholeplasma laidlawii B [Monck, M., Bloom, M., Lafleur, M., Lewis, R. N. A. H., McElhaney, R. N., & Cullis, P. R. (1992) Biochemistry 31, 10037-10043]. Since cholesterol increases hydrocarbon order in membranes, it was of interest to examine...
متن کاملAdsorption of mycoplasmavirus MV-L2 to Acholeplasma laidlawii: effects of changes in the acyl-chain composition of membrane lipids.
The enveloped mycoplasmavirus MV-L2 and its host Acholeplasma laidlawii JA1 were used to study the ways in which changes in the membrane lipid bilayer affect virus adsorption. The physical state of the membranes was altered by (i) using viruses and bacteria with different membrane lipid acyl-chain compositions, (ii) using incorporation of cholesterol, and (iii) changing the temperature. Adsorpt...
متن کاملLysis of Acholeplasma laidlawii by antibodies and complement.
Membranes of Acholeplasma laidlawii were used to determine the membrane components involved in immune lysis and to eventually detect enzymatic changes in the membrane components during this reaction. In a previous publication we reported that A. laidlawii can be killed by antibody and complement and that antibodies to membrane proteins are by far more effective than antibodies to membrane lipid...
